I suggest using an LM565 phase lock loop. Set the VCO frequency at the tone you want to check, use multiple switched trimpots if you want more than one frequency. Connect a microphone (probably need an amplifier) to the input, connect a voltmeter to the control voltage. The voltage will indicate how far it is off tune.
